Freeze-substitution is based on rapid freezing of tissues followed by solution (“substitution”) of ice at temperatures well below O°C – for the gentle fixation and dehydration of tissue. Freeze substitution is a process for low temperature dehydration and fixation of rapidly frozen cells that usually takes days to complete.
