While Lugol’s reagent will not reliably confirm amyloidity, surprisingly it greatly enhances visualization of Russula and Amanita subgenus Lepidella spores at high magnification (I haven’t tried this yet with non-amyloid spores). The trick is letting the solution under the cover slip to lose its background color (iodine dissipates/evaporates or gets destroyed photo-chemically), which typically occurs within half hour or so if you allow the slide to sit under bright ambient light. This is important because freshly made slides look blurry and rather dark at 100×. Mind you that spores remain stained permanently. Removing excess iodine from the slide will allow you to focus sharply and experience a much better contrast and low eye strain.
