Are you using congo red?
Thank you, Martin. Yes, it’s Congo Red in the last three pictures, and KOH. I first put a piece of tissue in CR, then wick (most of) that red liquid off from the slide and replace with a drop of KOH. Then press with a cover slip gently, and if the material does not spread nicely (which will typically happen), I remove the cover slide, go under the dissecting scope and use a pair of needles to divide the tissue and (often) remove irrelevant parts of it, then put another drop of KOH, press the cover slide and look again. And repeat the process as necessary. :)
