TEF-1-alpha sequencing results/discussion

A really dramatic change from uniformly yellow when young to a deep dark red at maturity. I’ll need to make sure to get out to my spots as soon as the red-pores start popping; to see if any young ones have yellow pores. A couple sunny days should get them going this week.

I completely agree with you guys. In these spot every year they come up very early collection is in the dehydrator. From last year in the same spot 205931.

Robert’s collection here is likely to be different from everything I had sequenced so far, granted that LSU cannot resolve some morphologically distinct red-pored boletes that are undoubtedly discrete species. A protein-coding locus should discriminate between them, but I am digressing…
Robert’s collection has an interesting feature — yellow pores when young and dark brick-red later in age, with no fading to orange or having a yellow rim. I don’t think I’ve seen that shade of red associated with any of our red-pored subvelutipes-like collections from 2016.
Dave, I think your 209319 = 251141, and the latter was sequenced. Its LSU was different from what we think is our best candidate for subvelutipes, obs 242312 / obs 242315.

of “red-pored” boletes of mine have featured pore surface yellow on buttons and/or a yellow marginal area several mm wide. Here’s an example obs 209319. Here’s one that I wish I had preserved obs 207985.

Is this something that occurs with B. subvelutipes? I have always noted red pores on buttons of what I call subvelutipes.

This observation seems interesting to me. Given the early occurrence, subvelutipes seems to make sense. Other than the yellow pores on the smallest button, these look like what I’d call subvelutipes.

Looking at my “subvelutipes” observations, there are are examples with yellow marginal area on the pores. obs 244662, obs 244300.

Cannot believe these things are out already. That’s very early, given your location!
These look rather different from other red-poreds we collected last year, with exception of some conifer-associated entities by Dave W. It’s interesting that the pores in the button are yellow – I recall that one or two of Dave’s vouchers had that feature, too. I agree that perhaps these are closer to what the true subvelutipes should look like. We should DNA barcode this collection.
