DNA sequencing results and discussion

> A clean and contiguous nrDNA sequence of 1687 bps was procured from this material. The nrITS portion consists of the last 707 bases; the nrLSU portion consists of the first 980 bases (through to the LR5 region). There are no ambiguous characters, so it’s a single haplotype for both loci.

> This nrLSU sequence is 99.9% similar to the corresponding fragment in the LSU of obs 210721 and 100% identical to the LSU of obs 279688. This empirical results suggests conspecificity of the three collections. For a discussion of pertinent BLASTn results see obs 210721.
As it has been stated by me a couple of time elsewhere on MO, the LSU sequencing effort of red-pored collections from the eastern/northeastern North America (SE Canada south to NC) so far identified 9 unique genotypes, representing at least as many morphological species, that are likely residents of the prolific Neoboletus-Sutorius association in the sense of Wu et al.1,2

> A BLASTn search of the nrITS sequence returned a hit list that is indicative of this taxon being part of a very morphologically and genetically diverse Neoboletus-Sutorius association. In particular, the top 4 hits are boletes from Canada collected by Renee Lebeuf and Yves Lamoureux (list sorted by max score):
> KY826093: Boletales sp. voucher B2842 = 706/707 = ~99.9% similar (no pic available)
> KY826098: Boletales sp. voucher B2858 = 705/708 = 99.6% similar (see pic at https://www.flickr.com/...)
> KY826193: Boletales sp. voucher RL042 = 691/692 = ~99.9% similar (see pic at https://www.flickr.com/...)
> KY826168: Boletales sp. voucher RL013; 690/692 = 99.7% similar (see pic at https://www.flickr.com/...)
Also, the last 542 characters of this nrITS sequence are a 99.9% match (1 base off) to the nrITS sequence of obs 279688.

Do you think the pore surface is originally brown and fading to dingy yellow, or the other way around? Have you preserved it?
