Used references: RET "Using both GenBank and my in-house database, I am able to get a 99.6% match for
MO290890. It is Amanita sp-44. In this case you need to trim the sequence so
that it doesn’t include an overlap with the ITS1F binding site. This is
explained in the posted document. The motifs showing the divisions of an SSU
fragment, the three parts of ITS are present. However the third character of
the LSU 5’ motif is missing; therefore you can only include the first two
characters of that motif in your sequence. I looked at the raw data file and
there is no indication of the third “T” of “TTTGACCTCAAATCA.” Unfortunately,
your signal quality dropped suddenly at a critical moment. So you should only
leave two “T’s” at the end of your sequence."

I see the match is better than first appeared.

There is a variable “AT” repeat present in ITS1. This should be discounted. Which means the the genetic distance between the sequence from this voucher and my two sequences is 0.4% in both cases

It looks there is a 2 character indel which is not consistent between any pair of the three species. Possibly it is two independently varying single character indels.
